if youre comparing brand to compounded, dose accuracy in the vial is the variable, not the molecule
#semaglutide 2025-01-31
- truncation_tru — £72 a month for compounded against what brand costs locally is why most people here use vials 10:05
- reship_rita — right so i held at 0.5 for 2 months and it was the best decision i made 12:18
- blank_run — FLOW was the kidney outcomes trial and it gets quoted in here for things it didnt measure 12:21
held there too
unrelated but does anyone actually run a five week step instead of four
not the same trial
2.4 is the ceiling
plateaus move
thats STEP 1
right so steady state is the bit people skip, judging a new dose after eight days tells you almost nothing
SELECT was cardiovascular outcomes in people with overweight and existing disease, not a weight trial
seven days ish
lost about 8kg on the low doses before i even got to 1, which surprised me
quick one moving my dose day by a day or two never did anything noticeable for me
14 months at 2.4 and the honest summary is diminishing returns after the first half
give it four weeks
£72 a month for compounded against what brand costs locally is why most people here use vials
wk 1-4 0.25mg
wk 5-10 0.50mg (held 2 extra weeks)
wk 11-14 1.00mg
wk 15-26 1.70mg (held, long)
wk 27+ 2.40mgthe long half life flattens the trough more than people expect, i never felt a day seven dip
STEP 1 ran to 68 weeks. thats the one everyone half remembers
half life is about a week so youre roughly four to five weeks to steady state on any new dose
is 2.4 the top or do people go past it
slower worked better
| Trial schedule | What most people here do | |
|---|---|---|
| Step interval | 4 weeks, fixed | 4-12 weeks, on symptoms |
| Holding | protocol deviation | normal and expected |
| Top dose | reached by design | often never reached |
| Coming down | not studied | common at maintenance |
did anybody find a difference between Homopeptide and pharmacy at the same 0.5
restarted after 25 months off at a low dose and the nausea arrived exactly like the first time, still working it out
mine faded too
i dont think the plateau is a dose problem most of the time, but i cant prove that
i stayed on 1.7 for 5 months and never went to 2.4, appetite was already where i needed it
is there a reason to hold at 1.7 rather than going to 2.4
coming back to this dose day is sunday for me only because thats when i remember, not because sunday matters
compounded and brand felt the same to me at 7.5, which is one person and nothing more
changed my mind on the four week rule, i think it depends entirely on how the current dose feels, ymmv
right so i held at 0.5 for 2 months and it was the best decision i made
wk 1-4 0.25mg
wk 5-10 0.50mg (held 2 extra weeks)
wk 11-14 1.00mg
wk 15-26 1.70mg (held, long)
wk 27+ 2.40mgFLOW was the kidney outcomes trial and it gets quoted in here for things it didnt measure
how long did the first 12kg take for people at the low doses
STEP is several trials, so saying STEP said x is usually wrong without a number after it
