#reconstitution 2025-10-05

Highlights from this day
  • tarpit_tam — both are defensible. what is not defensible is switching between them mid-vial and forgetting which one you are on 16:19
  • swirl_not_shake — which brings me to the real question. this one is a Hubei Jianxiang vial and the fill looks light to me 17:14
  • tarpit_tam — Janoshik for content and purity, yes. send a sealed vial, not one you already punctured 17:41
  • swirl_not_shake — last one i had done, for reference. came back on mass 18:43

why does everyone say 2ml specifically and not 1ml

II

benzyl alcohol is what makes it bacteriostatic. it is about 0.9% typically, i could be wrong

ok so 10mg in 1ml is 10mg/ml, so 2.5mg is 25 units. thats the one i use

GB

the vacuum will pull it in. let it. fighting it is how you get a face full of aerosol

II

a slightly collapsed cake is cosmetic. a puddle in the bottom is not

😂1🧊3

do i swirl for a specific length of time or just until its clear

how long after reconstituting before its actually dissolved

❤️3🎉13

aim the diluent at the glass wall, not at the powder. it goes in gentler

clean the whole way up

30 mg + 3.0 ml = 10 mg/ml

 2.5 mg -> 0.25 ml -> 25 u
 5.0 mg -> 0.50 ml -> 50 u
 7.5 mg -> 0.75 ml -> 75 u
10.0 mg -> 1.00 ml -> 100 u   <- a whole barrel, split it or use two pins
UU

i run 30mg into 1.5ml, so 20mg/ml, and take 12.5u for 2.5mg. half marks but a much smaller sting

TT

both are defensible. what is not defensible is switching between them mid-vial and forgetting which one you are on

UU

the powder itself takes up volume as well. 30mg of tirz is a visible cake

TT

a few tens of microlitres at that mass. that is below your measurement error, ignore it

TT

if you are working with 100mg and up it starts to matter

SN

which brings me to the real question. this one is a Hubei Jianxiang vial and the fill looks light to me

TT

cake height tells you almost nothing. change the lyophilisation cycle and you change the density

if you want to know the mass, weigh it or get it assayed. eyeballs do not do mass

TT

Janoshik for content and purity, yes. send a sealed vial, not one you already punctured

TT

different services, both fine. do not average their numbers, read them separately

SC

and PeptideMeter is where i look when i want a vendor history rather than one vial

SN

GL Biochem powder i had years ago dissolved slower than anything since and i never worked out why

TT

residual moisture and particle size. slow to dissolve is not the same as bad

SN

last one i had done, for reference. came back on mass
