Highlights from this day
  • coa_or_cope — the vial gets blamed for so much in this channel 23:11
  • area_percent — worked example, roughly, weekly half-life 23:19
  • blank_slate_bo — that explains the fortnight i had in may 23:21
  • luerlok — eleven weeks and the bot is prouder of me than my family 23:32

whats the longest anyone has stayed on one dose

Trial scheduleWhat most people here do

i jumped a rung once to catch up after a supply gap and it was a bad week. would not repeat

*week 2 not week 5

the trials escalated on a fixed schedule because a trial has to. you are not a trial, ill dig out the number

SD

with a roughly seven day half-life you are near steady state after about five weeks at a dose, from memory

SD

if you are asking whether to go up, the fact you are asking usually means not yet

LU

pick the day so the worst 48 hours land where you can afford them

MM

i went friday evening for two years so the rough saturday was mine and not my employers

CO

for me day 1 to 2 after. for others day 3. it varies and you will know within two doses

AP

the peak concentration for the weekly injectables is a day or two out, so day 1 to 2 is the common answer

AP

and move it later rather than earlier if you can. shortening the interval raises your average, lengthening it lowers it

AP

a small one, repeatedly, which is how people end up above the dose they think theyre on

LU

i drifted from sunday to thursday over about eight months and blamed the vial

MM

an alarm you cannot dismiss and a log you have to type into. thats it, theres no clever answer

CO

and take it late rather than skipping. a day late is nothing on a weekly half-life

AP

worked example, roughly, weekly half-life

on schedule, at steady state you sit around 2.0 "units"
of accumulated drug (see the doubling maths in #semaglutide)

skip one week entirely:
  day 7   2.0  -> no dose
  day 14 1.0
  day 21 0.5 + next dose

so one skipped week costs you about half your level and
takes ~3 weeks to build back. that is why a skipped dose
often shows up as a hungry fortnight, not a hungry day.
LU

and thats also why people who dose fortnightly on purpose are running a very different curve to a weekly one

AP

very different. same weekly average is not the same peak to trough, and appetite tracks trough more than average

CO

for cost mostly, and for maintenance sometimes. its in the maintenance conversation further down

VB

dose reminder for @luerlok: due today 19:00. current interval streak 11 weeks on schedule (personal best).

LU

eleven weeks and the bot is prouder of me than my family

MM

back. and the streak counter genuinely fixed my drift, which is embarrassing but true

AP

different question and yes, mostly. stretching the interval is the gentlest way to reduce exposure without changing what you draw

CO

thats basically the whole taper conversation and it starts below
