Highlights from this day
  • quiet.hours — an identical chromatogram on two different lots is the single biggest tell there is 20:57
  • panel_before_after — a certificate should name the method, the instrument, the column and the operator. most name none of those 22:06
  • taper_tess — coming back after 24 months, has the pinned reading guide changed 22:23
  • area_percent — one decimal versus two is usually just house style, not precision 22:34
  • area_percent — the font on this one changes halfway down, am i being paranoid 22:54

an identical chromatogram on two different lots is the single biggest tell there is

nobody can authenticate a certificate from a scan. you authenticate the material by testing it yourself

i keep every certificate in a dated folder and compare them. two suppliers had not changed their template since 2019

the related substances table is the interesting part. a row of "not detected" means they did not look hard

follow up a supplier certificate on its own is a starting point, not evidence. that is the whole reason the verification channel exists

residual solvents matter more than people think for anything that went through a lot of purification steps

🤝2📉8

net peptide is gross mass minus water minus counterion. that is why 11.4mg of powder can be a correct 10mg vial

i have never seen a research certificate with an endotoxin result on it. not once, take that with a pinch of salt

why do some certificates report to one decimal and some to two
an identical chromatogram on two different lots is the single biggest tell there is

the certificate from WWB is the one i use to teach this because it actually has a populated impurity table

PB

update from 13 months ago: started asking every supplier for water content and about half now include it

quick one no lot number, no certificate. a document you cannot tie to the vial in your hand is decoration

looks re-dated

TT

thats a tell

lot bought purity assay/label
A-2418 2025-03 99.1% 9.6 / 10mg
A-2601 2025-08 98.7% 9.8 / 10mg
B-0114 2026-01 99.3% 9.4 / 10mg
B-0329 2026-04 98.9% 9.9 / 10mg
TT

water content is why an honest 10mg vial assays at 9.1. karl fischer titration, and most certificates omit it

whats the minimum a certificate has to have before you take it seriously

the three way match is vial, certificate, invoice. if all three agree you have something

TT

sloppy not sinister

PB

while im here if you only read one line on a certificate, read the lot number. if you read two, read the assay

TT

purity is what fraction of the contents is the target. assay is how many mg are actually there. different questions

PB

a certificate should name the method, the instrument, the column and the operator. most name none of those

mixed fonts and inconsistent kerning in one document is worth a second look, your mileage will differ

gross mass on a jewellery scale plus the water content is enough to catch most underfills, anyway

TT

the honest summary is that a certificate tells you what the supplier says about a lot they chose to test

TT

coming back after 24 months, has the pinned reading guide changed

🤝1👍9🧊4

sterility and endotoxin are separate tests and almost nobody runs either on research material

AP

one decimal versus two is usually just house style, not precision

TT

if the certificate date precedes the lot date, ask. sometimes it is sloppiness and sometimes it is not
single wavelength detection can miss a co-eluting impurity that does not absorb there

AP

the font on this one changes halfway down, am i being paranoid

🧪10⚠️3

retention time shifts between runs are normal. relative retention time is the number to compare

no lot no certificate

DE

the paperwork can be perfect and the vial can still be light. weigh it

AA

a vial can be 99.4% pure and hold 8.1mg of a 10mg label. both numbers are true and only one of them is the one you care about
