Highlights from this day
  • swirl_not_shake — REDEFINE is the phase 3 programme, most of what gets quoted casually in here is earlier work, n of 1 obviously 16:03
  • vialkeeper — its an amylin analogue, different receptor family, so treating it as another GLP-1 is the first mistake, happy to be corrected 16:17
  • cagri_curious — stopped for 25 months and the fullness went within a fortnight of the last dose 16:46
  • cass_cagri — the reason nausea reads differently is timing, mine arrived on the second day rather than the first 18:44

🔥10🤝12

how many weeks before you noticed anything on monotherapy

VI

is there a reason people titrate this over more weeks than the others

🧊3

😂11🎉4

whats the recon people use, 40 in 0.5ml
stopped for 18 months and the fullness went within a fortnight of the last dose

SN

dropped the GLP-1 and stayed on this alone for 1 months, appetite held but weight barely moved

C1

combination GI was worse than either alone for me, which is not what i had read anywhere

🙏2
CH

is the nausea profile genuinely different or am i imagining it

C1

CagriSema is cagrilintide with semaglutide and REDEFINE is the trial programme around it, someone check my working

Cagrilintide monotherapy, dose ranging. The combination trials came later.
🧊4🤝2👀18

is the slower titration because of nausea or something else

💀11

*Medutest not the other one

noticed something around week 22 on monotherapy, well after i had given up expecting to

DE

i was sceptical about monotherapy and after 15 months im still sceptical, just less so

⚠️4
KF

lost 37lb over 2 months on the combination and i genuinely cannot apportion it

😂2🤝14
KF

dosing them on different days made no difference i could detect

monotherapy was underwhelming for me and i say that as someone who wanted it to work

DE

did the combination change your GI compared to the GLP-1 alone

the fullness effect hasnt faded over 13 months, unlike my semaglutide appetite curve

i keep the two on separate days so if something goes wrong i have a chance of knowing which

KF

monotherapy question, did appetite move for you without a GLP-1 alongside

did anyone add this to an existing semaglutide dose rather than starting both

VI

the slower titration in here is community habit as much as anything, the trials had their own schedule
nobody should copy my schedule, im describing it because people asked what a slow ramp looks like

SB

you cannot tell which of the two is doing what in a combination and i stopped pretending i could

the combination is the thing with trial data behind it, monotherapy in here is mostly us guessing

CC

how do people separate which of the two is doing what in a combination

the reason nausea reads differently is timing, mine arrived on the second day rather than the first

changed my mind on this, i started thinking amylin was the interesting bit and now i think its the pairing

fullness rather than nausea

VI

does amylin do something about fullness that the GLP-1 doesnt

VI

is the combination just the two dosed together or a single formulation

the fullness thing is the part people describe differently, food stops being interesting rather than repellent

GG

20 in 2ml gives 4mg/ml which is what i use, purely because the units come out round

KF

the nausea felt different rather than less, more fullness and less of the queasy wave, from memory

i titrated over twice as many weeks as i did on semaglutide and that was right for me
